Peptide and Protein Characterization Services (PPCS)

GENEPEP offers analysis of peptides and proteins originated from natural sources (organisms, tissues, body fluids), produced by synthesis or through recombinant strategies with:

Services provided:

Molecular mass (average, monoisotopic) measurement by MALDI-MS or / and ESI-MS;
Peptide sequencing by Edman chemistry or tandem mass spectrometry (MS / MS);
Protein characterization by enzymatic digestion, peptide fragment mass fingerprints;
De-novo sequencing (MS / MS) for the characterization of sequence tags;
Purification of the digest (s) by off-line HPLC or nanoflow on-line-LC-ESI-MS/MS, sequencing by Edman chemistry, and MS/MS;
Chemical treatments such as reduction / alkylation in solution or on solid supports;
Elucidation of post-translational modifications (PTMs) such as N-terminal cyclization, determination of the number of cysteine residues, etc;
Determination of the pairing of cysteine residues by enzymatic digestion and MS.

Detailed electronic report by e.mail in pdf or by fax on request. The report includes (i) a reference number, (ii) a brief description of the procedure, (iii) amino acid sequence(s), (iv) sequence coverage, (v) molecular mass (spectra are provided), (vi) HPLC profiles, (vii) protein identification, and (viii) database search if requested. Quality Control Services (QCS)

Genepep, through its QCS, offers the possibility to determine the purity and the identity of peptides and proteins. This includes peptides/proteins with non-natural amino acids, chemical modifications and post-translational modifications. QCS for peptides (natural or synthetic) and proteins (natural or recombinant), under standard procedures:

Measurement of the molecular mass (monisotopic or average) by MALDI-TOF-MS, by ESI-MS and / or by on line nanoLC-ESI-MS;
Determination of purity by MALDI-MS, ESI-MS and / or on line nanoLC-ESI-MS;
Control of purity by analytical reversed-phase (RP)-HPLC or nano RP-HPLC;
Highly detailed reports, validated by an expert in the field of mass spectrometry and liquid chromatography.
